Abstract

Flora diversity in Indonesia is unrivaled and famous than other countries especially orchids. Currently the population of black orchid (Coeloegyne pandurata)had decreased because demand of black orchids is increase but this is a problem that black orchid will existence in their habitat. These problems can be overcome with orchid propagation through in vitro culture. The study was conducted using a completely randomized design (CRD) with one factor which media with 20 replications. The aim of this study is to obtain information as preliminary data on the response time growthof black orchid seeds and compare Vacin and Went (VW) medium with organic medium. Variables studied in this research is a time to grow, number of explants, number of leaves and number of roots. The results of this research that respose of VW medium and organic medium can be used as a propagation medium for black orchid in tissue culture 5 week after initiation/germination (week-5). Media VW  has complete nutrients than organic medium.The result showed that the average number of explants (week-5) on mediumVW is 235.55 explants and  medium organic is 191 explants.The result at week – 16 showed that average number of medium VW is 308.85 explants and mediais 256.65 explants. Observations average number of leaves  showed that on mediumVW is 2.75 leaves and medium organic 2.05 leaves.
