Efek Pemberian Ekstrak Teh Hijau (Camellia Sinensis) terhadap Jumlah Sel Spermatid pada Toksisitas Testis Mencit yang Diinduksi Boraks

Abstract


ABSTRAK
Tumbuhan teh hijau berpotensi sebagai anti oksidan eksogen karena mengandung flavonoid yang dapat memutus reaksi berantai radikal bebas dengan melepaskan atom hidrogen dari gugus hidroksilnya. Penggunaan boraks berkepanjangan dapat memicu aktifitas radikal bebas pada tubuh. Radikal bebas dapat merusak DNA melalui peroksidasi lipid yang terdapat di membrane sel spermatid pada organ testis. Tujuan penelitian mengetahui efek pemberian ekstrak teh hijau terhadap jumlah sel spermatid mencit jantan strain BALB/c yang diinduksi boraks. Penelitian menggunakan rancangan posttest only control group design dengan 35 ekor mencit jantan Strain BALB/c yang dibagi dalam 6 kelompok secara acak. Hari ke-36 dilakukan terminasi jaringan testis untuk dibuat sediaan preparat histologi. Pewarnaan preparat menggunakan pengecatan H & E kemudian diamati dengan perbesaran 400x. Gambaran histopatologi sel spermatid dinilai berdasarkan rerata jumlah sel spermatid dini dan matur pada 3 lapang pandang. Data dianalisis dengan One Way Annova dan dilanjutkan dengan uji Poshoc menggunakan LSD. Hasil rerata K-I, K-II, K-III, K-IV, K-V, dan K-VI adalah 123.22±20.10, 84.67±3.30, 132.8±6.53, 139.55±9.43, 138.89±8.12, dan 134.22±8.52 secara berurutan. Uji One Way Annova 0,000 (p<0,005) menunjukan terdapat perbedaan bermakna. Uji Posthoc dengan LSD menunjukan terdapat perbedaan bermakna antara: K-II dan K-I, K-III, K-IV, K-V, K-V; K-I dan K-IV, KV. Pemberian ekstrak teh hijau dapat meningkatkan jumlah sel spermatid mencit jantan Strain BALB/c yang diinduksi boraks.

ABSTRACT
Green tea has the potential as an exogenous antioxidant because it contains flavonoids which can break free chain reactions by releasing hydrogen atoms from their hydroxyl groups. Prolonged use of borax can trigger free radical activity in the body. Free radicals can damage DNA through lipid peroxidation found in spermatid cell membranes in testis. The study aims to determine the effect of green tea extract on the number of spermatid cells in male mice BALB/c strain induced by borax. In this study using a posttest only control group design. 35 male BALB/c mice were divided into six groups randomly. On day 36, the testicular tissues were taken for H&E stained histological preparation and observed under microscope with 400x magnification. The number of spermatid cells were assessed based on the average number of early and mature spermatid cells in three visual fields. Then the data were analyzed using One Way Annova and continued with Poshoc test using LSD.The mean of spermatid cell number of K-I, K- II, K-III, K-IV, K-V, K-VI was 123.22±20.10, 84.67±3.30,132.83±6.53, 139.55±9.43, 138.89±8.12, 134.22±8.52 respectively. One Way Annova resulted in p<0.005 showing that was a significant difference. The result of Posthoc test with LSD showing that there was a significant difference between: K-II andK-I, K-III, K-IV, K-V, K-VI; K-I and K-IV, K-V. Green tea extract could increase the number of spermatid cells in boric acid induced testicular toxicity in mice.
